Biochemical and physiological responses of sugarcane cultivars to soil water deficiencies Scientia Agricola

For sugarcane (Saccharum spp.) crops, the effects of an environmental stress, especially water deficiency, may cause severe productivity reduction, inferring negatively in the sugarcane industry. The tolerance of two sugarcane cultivars to a lack of water was made by analyzing the levels of the osmoprotectors, trehalose and free proline, and the biometrical variables of their initial growth. Biochemical and physiological responses of the cultivars, when subjected to water stress, were assayed to determine how these plants tolerate drought. Changes in trehalose content of baker's yeast as affected by octanoic acid Scientia Agricola

Octanoic acid inhibited ethanolic fermentation by Saccharomyces cerevisiae (bakers yeast) and the trehalose accumulation, however did not affect the endogenous degradation of trehalose. This inhibition may be explained by the binding of octanoic acid to hexokinase or other proteins of plasma membrane because they are not necessary for endogenous fermentation. The degradation of trehalose may be due to an activation of trehalase.

Screening for such biotherapeutic agents is commonly performed by in vitro assays simulating gastrointestinal environment to determine the ability to survive in the digestive tract. Less than 0.5% of total water in the world is available for human consumption and agriculture. The major part of the world's water is saline and salinity in soils interferes in germination of seeds and the posterior development of the plant. The fermentation of yeast reserve carbohydrates, glycogen and trehalose is a procedure to increase protein level of yeast cells and improve ethanol production. This work studied on the degradation kinetics of glycogen and trehalose carried out with two industrial strains of Saccharomyces cerevisiae (PE-2 and SA-1) and the effect of different temperatures (38º, 40º, 42º and 44ºC) on degradation rate. Endogenous fermentation was carried out in a yeast suspension at 20% (w/v) based on wet weight, suspended in fermented media with 3.0 to 4.5 % (v/v) of ethanol. Shared control of maltose and trehalose utilization in Candida utilis BJMBR

Trehalose biosynthesis and its hydrolysis have been extensively studied in yeast, but few reports have addressed the catabolism of exogenously supplied trehalose. Here we report the catabolism of exogenous trehalose by Candida utilis. In contrast to the biphasic growth in glucose, the growth of C. utilis in a mineral medium with trehalose as the sole carbon and energy source is aerobic and exhibits the Kluyver effect. Trehalose is transported into the cell by an inducible trehalose transporter (K M of 8 mM and V MAX of 1.8 µmol trehalose min-1 mg cell (dry weight)-1. Thermostabilisation of human serum butyrylcholinesterase for detection of its inhibitors in water and biological fluids BABT

The ability of gelatine-trehalose to convert the normally fragile, dry human serum BChE into a thermostable enzyme and its use in the detection of cholinesterase inhibitors in water and biological fluids is described. Gelatine or trehalose alone is unable to protect the dry enzyme against exposure to high temperature, while a combination of gelatine and trehalose were able to protect the enzyme activity against prolonged exposure to temperature as high as +50°C. A method for rapid, simple and inexpensive means of screening for cholinesterase inhibitors such as carbamates and organophosphates in water, vegetables and human blood has been developed.

Trehalose accumulation, invertase activity and physiological characteristics of yeasts isolated from 24 h fermentative cycles during the production of artisanal Brazilian cachaça BJM

Trehalose accumulation, invertase activity and physiological characteristics of 86 yeast isolates from short fermentative cycles during the production of cachaça in three artisanal distilleries of the State of Minas Gerais were studied. Among these isolates, 70% were able to grow at temperatures between 40 and 42ºC. Only Saccharomyces cerevisiae isolates were able to grow over 40ºC. Lower temperatures (<40ºC) favoured the growth of yeasts such as Candida parapsilosis-like, C. maltosa-like, Kloeckera japonica, S. exiguus and C. bombicola-like.
